It's basically "Ancient Architects".
The Mazone lived on Earth long ago and migrated to another system for some reason. Or the humans evolved from Mazone that were left behind, or were animals that the Mazone had genetically touched.
For what we watched the Mazone now is very different, with their plant-like bodies. They may have had to change to survive their travels, except the rulers, that remained with human-like bodies.
Or the other way around.
Anyway, it's implied that the Mazone is linked to the humans and for me is really shitty for Harlock to not kill her just because of this doubt. What's the point in all he did against the Mazone? If he recognizes Lafresia as human then shouldn't he let her go to Earth keeping with his "rule" to not kill and interfere with humans?
